      [目的]探讨脾虚型溃疡性结肠炎(ulcerative colitis,UC)患者的肠道菌群特征。[方法]收集2019年1月1日至2019年3月31日于浙江中医药大学附属第二医院、浙江省中医院、浙江省人民医院以及浙江大学附属第二医院门诊及住院的14例脾虚型UC患者、16例其他证型UC患者的粪便样本,同时收集同时间段至浙江中医药大学附属第二医院体检的8例健康者的粪便样本,提取三组粪便样本中菌群DNA,根据16S V3~V4区设计引物进行扩增,通过Illumina Miseq平台进行测序,以生物信息分析软件对测序结果进行分析,比较三组样本菌群组成和结构,分析脾虚型UC患者的肠道菌群特征。[结果]门分类水平上其他证型UC组中变形杆菌门的相对丰度显著高于健康人群组(P<0.01)。属分类水平上,脾虚型UC组和其他证型UC组中分枝杆菌属、包西氏菌属、丁酸单胞菌属的相对丰度显著低于健康人群组(P<0.05),而埃希氏菌属/志贺氏菌属、假单胞菌属的相对丰度明显高于健康人群组(P<0.05)。此外,代尔夫特菌属的相对丰度在脾虚型UC组中明显低于其他证型UC组和健康人群组(P<0.05)。[结论]脾虚型UC患者的肠道菌群特征与其他证型UC患者及健康人群相比存在一定的差异,代尔夫特菌属明显减少可能是脾虚型UC患者肠道菌群的特征性变化。

      [Objective] To explore the characteristics of intestinal flora in patients with ulcerative colitis(UC) of spleen-deficiency type. [Methods] The fecal samples of 14 UC patients of spleen-deficiency type and 16 UC patients of other syndromes were collected from January 1 to March 31, 2019 in outpatient and inpatient departments of the Second Affiliated Hospital of Zhejiang Chinese Medical University, Traditional Chinese Medicine Hospital of Zhejiang Province, Zhejiang Provincial People's Hospital and the Second Affiliated Hospital of Zhejiang University. Meanwhile, the fecal samples of 8 healthy people who went to the Second Affiliated Hospital of Zhejiang Chinese Medical University for physical examination in the same period were collected. The total bacterial DNA was amplified according to the 16S V3~V4 region, and sequencing was performed on the Illumina MiSeq platform. The sequencing results were analyzed by bioinformatics analysis software. The flora composition and structure of the samples from three groups were compared. [Results] At the classification level of phylum, compared with the healthy group, the relative abundance of Proteobacteria in other syndromes group was significantly increased(P<0.01); at the classification level of genus, compared with healthy group, the relative abundance of Mycobacterium, Bosea, Butyricimonas in the spleen-deficiency group and other syndrome group was significantly decreased(P<0.05), while the relative abundance of Escherichia/Shigella and Pseudomonas was significantly increased(P<0.05);furthermore, the relative abundance of Delftia genus in the spleen-deficiency group was significantly decreased compared with other syndromes group and healthy group (P<0.05). [Conclusion] The intestinal flora characteristics of UC patients with spleen-deficiency are different from those of UC patients with other syndromes and the healthy group. The significant decrease of Delftia may be a characteristic change of intestinal flora in UC patients of spleen-deficiency type.
