      [目的]观察电针(electroacupuncture,EA)对吗啡耐受(morphine tolerance,MT)模型大鼠机械性痛阈(mechanical paw-withdrawal threshold,MPWT)、脊髓背角NOD样受体蛋白3(Nod-like receptor protein 3,NLRP3)炎症小体及其炎症因子表达的调控。[方法]将18只健康雌性SD大鼠按完全随机法分为3组,包括空白组、模型组和EA组,每组6只。模型组和EA组大鼠于鞘内连续注射吗啡7d,制备MT模型,分别于开始造模后第1、3、5、7天检测大鼠左足MPWT。建模成功后,模型组和EA组继续予鞘内注射吗啡,连续7d,EA组在予鞘内吗啡注射后0.5h,予双侧“足三里”“昆仑”穴位EA治疗,1次/d,连续7d,各组分别于建模成功后第1、3、5、7天检测左足MPWT。以Western blot法检测大鼠脊髓背角NLRP3炎症小体、白细胞介素-18(interleukin-18,IL-18)、白细胞介素-1β(interleukin-1β,IL-1β)表达。[结果]与空白组比较,模型组大鼠注射吗啡后第1、3、5天MPWT显著升高(均P<0.01),但注射吗啡7d后降低至基础水平,与空白组比较差异无统计学意义(P>0.05),说明MT模型造模成功。与模型组比较,EA组大鼠第3、5、7天的MPWT升高(P<0.01)。与空白组比较,模型组大鼠脊髓背角中NLRP3炎症小体、IL-18和IL-1β表达增高(均P<0.01);与模型组比较,EA组大鼠脊髓背角中NLRP3炎症小体、IL-18和IL-1β表达减低(均P<0.01)。[结论]EA能缓解吗啡耐受效应,其机制可能与下调脊髓背角NLRP3炎症小体、IL-18和IL-1β的表达有关。

      [Objective] To observe the effect of electroacupuncture(EA) on mechanical paw-withdrawal threshold(MPWT) and its regulation of Nod-like receptor protein 3(NLRP3) inflammatory body and inflammatory factors in spinal dorsal horn in morphine tolerance(MT) rats.[Methods] Eighteen SD rats were randomized into blank group, model group and EA group, 6 rats in each group. MT models were set up by continuously injecting morphine intrathecally for 7 days, and MPWT on the left was detected on d1, d3, d5, and d7. After successful modeling, rats in model group and EA group were continued to be injected intrathecally with morphine for 7 days. The EA group was given morphine intrathecally, and bilateral "Zusanli" and "Kunlun" acupoints were stimulated with EA 0.5h after injection, once per day for 7 days. MPWT on the left was detected at d1, d3, d5, and d7. The contents of NLRP3 inflammatory body, interleukin-18(IL-18) and interleukin-1β(IL-1β) in the spinal dorsal horn were detected by Western blot.[Results] Compared with blank group, MPWT of model group significantly increased after d1,d3,d5 of morphine injection(P<0.01), and decreased to the basic level after 7 days of morphine injection, compared with blank group, there was no significant difference(P>0.05), indicating that the MT model was successful. Compared with model group, the MPWT at d3, d5 and d7 of EA group increased after MT model set up(P<0.01). Compared with blank group, the contents of NLRP3 inflammatory body, IL-18 and IL-1β in the spinal dorsal horn of model group increased(all P<0.01); compared with model group, the contents of NLRP3 inflammatory body, IL-18, and IL-1β in the spinal dorsal horn of EA group decreased(all P<0.01).[Conclusion]EA relieves MT effect, and its mechanism may be related to down-regulation of NLRP3 inflammatory body, IL-18 and IL-1β expression in dorsal horn of spinal cord.
