      [目的]观察培补二天膏对老年自发性高血压大鼠(spontaneously hypertensive rats,SHR)血压、炎性细胞因子和心脏重构的影响,探讨其降压疗效及改善心脏重构的可能机制。[方法]选取12月龄雄性SHR共50只,基础血压测定达标后随机分为5组:阴性对照组,膏方低、中、高剂量组,厄贝沙坦组;另选取同月龄Wistar Kyoto(WKY)大鼠9只作为WKY组。膏方组分别予0.58g/(kg·d)、1.15g/(kg·d)和3.46g/(kg·d)培补二天膏膏方灌胃,厄贝沙坦组以厄贝沙坦10.13mg/(kg·d)灌胃,阴性对照组及WKY组予温蒸馏水灌胃,各组灌胃均为4周。采取尾动脉搏动测量法,测定给药后每周大鼠尾动脉收缩压(systolic blood pressure,SBP),4周后检测各组大鼠血清白介素-1β(interleukin-1β,IL-1β)、IL-2、IL-6、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)水平,同时测定各组大鼠心脏重量指数(heart mass index,HMI)。苏木素-伊红(hematoxylin-eosin,HE)染色后镜下观察心室肌切片的病理学形态改变。[结果]灌胃4周后,阴性对照组血压高于WKY组(P<0.01);膏方中剂量及高剂量组血压较阴性对照组降低(P<0.01)。阴性对照组血清IL-1β、IL-2、IL-6、TNF-α水平均高于WKY组(P<0.01、P<0.05);膏方各剂量组SHR血清IL-1β、IL-2、IL-6、TNF-α水平均低于阴性对照组(P<0.01)。灌胃4周后,阴性对照组HMI高于WKY组(P<0.01);膏方高剂量组及厄贝沙坦组HMI低于阴性对照组(P<0.05)。镜下观察发现,膏方高剂量组及厄贝沙坦组心肌细胞肥大减轻,间质炎性细胞浸润较阴性对照组及膏方低剂量组明显减少。[结论]中、高剂量培补二天膏降低SHR血压的疗效确切,高剂量还能够改善SHR心脏重构。培补二天膏的降压疗效可能与炎性细胞因子的降低有关,并可通过降压及降低炎性细胞因子的作用改善心脏重构。

      [Objective] To study the effects of Peibu Ertian Paste on blood pressure, inflammatory cytokines and cardiac remodeling in elderly spontaneously hypertensive rats(SHR), and explore its antihypertensive effect and the possible mechanism of cardiac remodeling.[Methods] Fifty male 12-month aged SHRs were randomly divided into 5 groups, 7 in negative control group and 9 in paste low,medium and high dose groups respectively, and 10 in Irbesartan group; nine 12-month aged Wistar Kyoto(WKY) rats were used as WKY group. SHRs in paste low, medium and high dose groups were fed with 0.58g/(kg·d), 1.15g/(kg·d), 3.46g/(kg·d) paste separately, the Irbesartan group was fed with Irbesartan 10.13mg/(kg·d), rats in negative control group and WKY group were fed with warm distilled water. The tail artery pulsation measurement method was used to measure the systolic blood pressure(SBP) of the tail artery of the rats before and after administration, and the serum levels of interleukin-1β(IL-1β), IL-2, IL-6 and tumor necrosis factor-α(TNF-α) were detected after 4 weeks of gavage. The heart mass index(HMI) of each group was measured. Ventricular muscle sections were stained by hematoxylia-eosin(HE) staining and pathological changes were observed.[Results] After administration, SBP of negative control group was higher than that of the WKY group(P<0.01).SBP of paste medium and high dose group was lower than that of negative control group(P<0.01). Serum IL-1β, IL-2, IL-6 and TNF-α levels of negative control group were higher than those in WKY group(P<0.01, P<0.05).The serum IL-1β,IL-2,IL-6,TNF-α level of paste groups was lower than that of negative control group(P<0.01). After four weeks of administra-tion, HMI of negative control group was higher than that of WKY group(P<0.01). HMI of paste high dose group and Irbesartan group were lower than those of negative control group(P<0.05).In paste high dose group and Irbesartan group, the myocardial cell hypertrophy was alleviated, and the interstitial inflammatory cell infiltration was significantly reduced compared with negative control group and paste low dose group.[Conclusion] The effect of medium and high dose Peibu Ertian Paste on reducing SHR blood pressure is exact. The antihypertensive effect of Peibu Ertian Paste may be related to the reduction of inflammatory cytokines, and its antihypertensive and inflammatory cytokine-reducing effects improve SHR cardiac remodeling.
