      目的 探讨化痰行瘀汤对四氯化碳(CCL4)诱导肝纤维化大鼠的治疗作用。方法 将72只雄性成年SD大鼠随机分为正常组、模型组、化痰行瘀汤高、中、低剂量组及阳性对照组。除正常组外,其余各组均用腹腔注射CCL4制备肝纤维化模型。各治疗组于第6周开始给药,化痰行瘀汤按高、中、低量灌胃,阳性对照组给与复方鳖甲软肝片灌胃,正常组、模型组给予等容量生理盐水灌胃,至第10周结束。观察各组大鼠肝脏病理学变化;测定血清ALT、AST、ALP及Ⅲ型前胶原(PCⅢ)、Ⅳ型胶原(Ⅳ-C)、透明质酸(HA)、层黏连蛋白(LN)含量的变化,并测定肝组织中脂质过氧化相关指标过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)和丙二醛(MDA)等。结果 与正常组比较,模型组大鼠肝脾指数升高,血清ALT、AST、ALP及PCⅢ、Ⅳ-C、HA、LN显著升高,肝组织中CAT、GSH-Px及SOD活性显著降低,MDA显著升高(P均<0.01)。与模型组比较,化痰行瘀汤高剂量组和复方鳖甲软肝片组大鼠肝脾指数降低,血清ALT、AST、ALP及PCⅢ、Ⅳ-C、HA、LN降低,肝组织CAT、GSH-Px及SOD活性升高,MDA下降(P均<0.01);化痰行瘀汤中剂量组肝脾脏指数下降(P<0.05),血清ALT、ALP下降(P<0.01)、AST 下降(P<0.05),PCⅢ、IV-C、HA、LN均明显下降(P<0.05),肝组织中CAT、GSH-Px及SOD水平升高(P<0.05),MDA水平下降(P<0.05)。化痰行瘀汤低剂量组与模型组比较各项指标均无明显差异。结论 化痰行瘀汤可以减轻肝纤维化并具有抗脂质过氧化的作用。

      Objective To investigate the therapeutic effect of Huatan Xingyu Decoction on liver fibrosis induced by carbon tetrachloride (CCL4) in rats. Methods 72 adult male SD rats were randomly divided into normal group, model group, high, medium and low dose group of Huatan Xingyu Decoction and positive control group. Except for the normal group, the other groups were intraperitoneally injected with CCL4 to prepare liver fibrosis model. Each treatment group began to administer medicine at the 6th week. The Huatan Xingyu Decoction was administered in high, medium and low doses. The positive control group was administered with Compound Biejia Ruangan Tablet. The normal group and the model group were administered with normal saline at the same volume until the end of the 10th week. The changes of serum ALT, AST, ALP and type III procollagen (PC III), type IV collagen (IV-C), hyaluronic acid (HA), laminin (LN) were observed, and the related indexes of lipid peroxidation, catalase (CAT), glutathione peroxidase (GSH-Px), superoxide dismutase (SOD) and malondialdehyde (MDA) were determined. Results Compared with the normal group, the liver and spleen index of the model group increased, the serum ALT, AST, ALP and PC III, IV-C, HA, LN increased significantly, the activities of CAT, GSH-Px and SOD decreased significantly, and the MDA increased significantly (P < 0.01). Compared with the model group, the liver and spleen index of rats in the high dose group of Huatan Xingyu Decoction and the compound Biejia Ruangan Tablet group decreased, the serum ALT, AST, ALP and PC III, IV-C, HA, LN decreased, the activities of CAT, GSH-Px and SOD in liver tissue increased, and MDA decreased (all < 0.01); the liver and spleen index of the middle dose group of Huatan Xingyu Decoction decreased (P < 0.05), the serum ALT, ALP < 0.01, AST decreased (P < 0.01). PCIII, IV-C, HA and LN decreased significantly (P < 0.05), while the levels of CAT, GSH-Px and SOD increased (P < 0.05) and MDA decreased (P < 0.05). There was no significant difference between the low-dose group and the model group in the indexes of Huatan Xingyu Decoction. Conclusion Huatan Xingyu Decoction can alleviate liver fibrosis and has anti-lipid peroxidation effect.
