[目的]研究白及对阿司匹林诱导的胃溃疡的治疗作用。[方法]将48只雄性Wistar大鼠随机分为正常组、模型组、白及高、中、低剂量组和阳性对照组。除正常组外,余各组大鼠均以阿司匹林溶液灌胃建立胃溃疡模型。8周后,阳性对照组以奥美拉唑灌胃,白及高、中、低剂量组以白及混悬液灌胃治疗。4周后评价各组大鼠胃溃疡指数,HE染色观察胃组织病理变化,ELISA法检测大鼠血清肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-6(interleukin-6,IL-6)及胃黏膜前列腺素E2(prostaglandin E2,PGE2)、内皮素(endothelin,ET)的表达水平;荧光定量PCR、Western blot分别检测胃黏膜环氧化酶-1(cyclooxygenase-1,COX-1)、环氧化酶-2(cyclooxygenase-2,COX-2)基因和蛋白表达。[结果]与正常组比较,模型组大鼠溃疡指数增加(P<0.01),血清TNF-α、IL-6表达增加(P<0.01),胃黏膜PGE2表达减少(P<0.01)、ET表达增加(P<0.01),COX-1基因和蛋白表达减少(P<0.01)、COX-2基因和蛋白表达增加(P<0.01,P<0.05),胃组织病理损伤明显。与模型组比较,白及各剂量组溃疡指数降低(P<0.01,P<0.05),血清TNF-α、IL-6表达减少(P<0.01,P<0.05),胃黏膜PGE2增加(P<0.01,P<0.05)、ET减少(P<0.01,P<0.05),COX-1基因和蛋白表达均增加(P<0.01,P<0.05)、COX-2基因和蛋白表达均减少(P<0.01,P<0.05),胃黏膜组织病理损伤减轻,其中,白及高剂量组的治疗效果最明显。[结论]白及能够有效减轻阿司匹林对大鼠胃黏膜的损伤,该作用与降低血清TNF-α与IL-6,调节胃黏膜COX-1、COX-2的表达水平,从而升高PGE2、减少ET有关。 |

[Objective] To study the therapeutic effect of Bletilla striata on gastric ulcer induced by aspirin. [Methods]Forty-eight rats were randomly divided into normal group, model group, three different dose of Bletilla striata suspension groups and positive control group. Except normal group, other groups were intragastric administrated with aspirin. After eight weeks, omeprazole and different doses of Bletilla striata suspension were administered respectively. Four weeks later, the gastric ulcer index of each group was evaluated and the expression of tumor necrosis factor-α(TNF-α), interleukin-6(IL-6) in serum and prostaglandin E2(PGE2), endothelin(ET) in gastric mucosal were detected by enzyme linked immunosorbent assay(ELISA). The expressions of cyclooxygenase-1(COX-1) and cyclooxygenase-2(COX-2) gene and protein expression in gastric mucosa were detected by fluorescence quantitative PCR and Western blot. [Results] Compared with normal group, the gastric ulcer index of model group increased(P<0.01), serum TNF-α and IL-6 increased(P<0.01), gastric mucosa PGE2 decreased(P<0.01), ET increased(P<0.01), the expression of COX-1 reduced(P<0.01), and COX-2 increased(P<0.01, P<0.05), the stomach tissue pathological damage was obvious. Compared with model group, the ulcer index decreased in each Bletilla striata suspension group(P<0.01, P<0.05), serum TNF-α, IL-6 reduced(P<0.01, P<0.05), gastric mucosal PGE2 increased(P<0.01, P<0.05), ET decreased(P<0.01, P<0.05), the expression of COX-1 increased (P<0.01, P<0.05), and COX-2 reduced (P<0.01, P<0.05), pathological injury reduced. Among them, the highest dose of Bletilla striata suspension has the best therapeutic effect. [Conclusion] Bletilla striata can effectively treat the gastric mucosal injury of aspirin in rats, which can reduce the expression of TNF-α and IL-6 and regulate the expression of COX-1 and COX-2 in gastric tissues, thereby increasing PGE2 and decreasing ET. |
