[目的]观察消瘀泄浊饮对慢性环孢素肾病大鼠肾素(renin)、血管紧张素Ⅱ(angiotensinⅡ,AngⅡ)、转化生长因子-β1(transforming growth factor-β1,TGF-β1)的影响。[方法]将40只雄性SD大鼠随机分为空白对照组、模型组、消瘀泄浊饮组、洛汀新(盐酸贝那普利)组各10只。采用环孢素灌胃及低盐饮食建模,治疗4周后检测大鼠血清肌酐(serum creatinine,Scr)、尿素氮(blood urea nitrogen,BUN)、尿肌酐、24小时尿蛋白总量,并计算肌酐清除率(creatinine clearance,Ccr),用苏木精-伊红染色法(hematoxylin-eosin staining,HE)观察大鼠的肾脏组织病理学改变,免疫组化法观察肾脏组织IV型胶原(collagen-Ⅳ,Col-Ⅳ)表达情况;酶联免疫吸附(enzyme-linked immunosorbentassay,ELISA)法检测血清及肾组织Renin水平,实时荧光定量PCR(quantitative real-time PCR,qPCR)检测肾脏组织AngII、TGF-β1mRNA表达情况。[结果]相比于模型组,洛汀新组及消瘀泄浊饮组大鼠Scr、Bun、Ccr及24小时尿蛋白明显改善,血清肾素水平下降,肾组织AngⅡ、TGF-β1mRNA及肾素、Col-Ⅳ表达下调,肾脏病理纤维化程度明显降低,消瘀泄浊饮组大鼠死亡率低于洛汀新组和模型组。[结论]消瘀泄浊饮可通过下调Renin、AngⅡ表达、抑制TGF-β信号转导,延缓慢性环孢素肾病的进展。 |

[Objective] To observe the effect of Xiaoyu Xiezhuo Decoction on renin, Ang, TGF-β1 in rats with chronic cyclosporine(CsA) nephropathy (CCN). [Methods] Forty male SD rats were divided into 4 groups: Control group, model group, Xiaoyu Xiezhuo group, benazepril group. The rats models were induced by CsA with low salt diet. After 4-week’s treatment, blood urea nitrogen(BUN),serum creatnine(Scr),urine creatnine and 24 hours urine protein(24hUpro) were detected,and creatnine clearance rate(Ccr) was calculated. Pathological changes of renal tissue were observed by HE staining. The expression of Col-IV in renal tissue was observed by immunohistochemistry. The renin levels in serum and renal tissue were detected by ELISA. The expressions of AngⅡ and TGF-β1mRNA were detected by qPCR. [Result]Compared with model group,the levels of BUN,Scr,24h Upro and Ccr in Xiaoyu Xiezhuo group and benazepril group were significantly better. The levels of renin in serum were decreased. The expressions of AngⅡ,TGF-β1 mRNA and renin, ColIV in renal tissue were significantly down-regulated. The degree of renal interstitial fibrosis decreased obviously. The death rate of rats in Xiaoyu Xiezhuo Decoction group was lower than that of thebenazepril group and model group. [Conclusion] Xiaoyu Xiezhuo Decoction can delay the progression of CCN via inhibiting TGF-β signal transduction by reducing the expression of Renin, AngⅡ. |
