      [目的]通过慢性束缚应激(chronic restraint stress,CRS)建立小鼠抑郁症模型,探究天麻水煎剂对小鼠抑郁样行为和神经递质5-羟色胺(5-hydroxytryptamine,5-HT)含量的影响。[方法]32只C57BL/6小鼠随机分为正常对照组、模型组、阳性药物组和天麻组,每组8只。除正常对照组外,其他小鼠均进行连续21d、每天6h的束缚刺激,另外阳性药物组每天予10mg/kg盐酸氟西汀灌胃,天麻组每天予1g/kg的天麻水煎剂灌胃。记录小鼠体重变化,并进行悬尾试验、强迫游泳试验、旷场试验,造模结束后检测小鼠肝功能指标谷丙转氨酶(alanine aminotransferase,ALT)和谷草转氨酶(aspartate aminotransferase,AST)的水平,并测定各组小鼠血清中肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-1β(interleukin-1β,IL-1β)和脑组织TNF-α、IL-1β、5-HT含量。[结果]天麻水煎剂灌胃对CRS小鼠血清ALT和AST水平无显著影响(P>0.05),表明在有效剂量下天麻水煎剂不具有肝毒性。与正常对照组比较,模型组小鼠表现出明显的抑郁样行为,糖水偏好指数降低(P<0.01),悬尾不动和强迫游泳不动时间增加(P<0.01),旷场试验中的中心区域移动距离和时间减少(P<0.01)。与模型组比较,天麻组小鼠糖水偏好指数显著增加(P<0.01),悬尾不动和强迫游泳不动时间显著降低(P<0.05),旷场试验中的中心区域移动距离和时间显著增加(P<0.05,P<0.01)。进一步检测显示,天麻组小鼠血清和脑组织中TNF-α和IL-1β含量显著降低(P<0.05,P<0.01),脑组织5-HT含量显著升高(P<0.05)。[结论]天麻水煎剂能够通过降低促炎细胞因子TNF-α和IL-1β含量,缓解炎症对中枢神经系统的压力,同时升高神经递质5-HT在海马区的含量,刺激5-HT能系统,缓解CRS小鼠的抑郁行为。

      [Objective] To establish mice model of depression by chronic restraint stress(CRS), and to investigate the effects of the water decoction of Gastrodia elata on depression-like behavior and 5-hydroxytryptamine(5-HT) content in modeled mice.[Methods] Thirty-two C57BL/6 mice were randomly divided into normal control group, model group, positive drug group and water decoction of Gastrodia elata group, with 8 rats in each group. Except for normal control group,the mice of other groups were subjected to restraint stimulation for 6 hours every day for 21 days, positive drug group was given fluoxetine hydrochloride (10mg/kg) and water decoction of Gastrodia elata group was given water decoction of Gastrodia elata(1g/kg) and the body weight changes of the mice were recorded. The tail suspension test, forced swimming test and open field test were carried out, and the indexes of liver function alanine aminotransferase(ALT), aspartate aminotransfease(AST),serum tumor necrosis factor-α(TNF-α) and interleukin-1β (IL-1β) content and brain tissue TNF-α, IL-1β, 5-HT content were measured after the end of modeling.[Results] There was no significant effect of water decoction of Gastrodia elata on serum ALT and AST level in CRS mice (P>0.05), which indicated that water decoction of Gastrodia elata had no hepatotoxicity in effective dose.Compared with normal control group, the mice in model group showed obvious depression-like behavior, the preference index of mice to sugar water decreased(P<0.01), the perfusion time and forced swimming time of CRS mice increased(P<0.01),the distance and time of movement to the central region were decreased(P<0.01).Compare with model group, the preference index of mice to sugar water was significantly increased(P<0.01),the perfusion time and forced swimming time of water decoction of Gastrodia elata group significantly decreased(P<0.05),and the distance and time of movement to the central region increased significantly(P<0.05,P<0.01).The TNF-α and IL-1β content in serum and brain tissue was significantly reduced(P<0.05,P<0.01), while the 5-HT content in brain tissue was significantly increased in water decoction of Gastrodia elata group(P<0.05). [Conclusion] Water decoction of Gastrodia elata can reduce the content of inflammatory cytokines TNF-α and IL-1β, relieve the pressure of inflammation on the central nervous system, increase the content of neurotransmitter 5-HT in hippocampus, and stimulate serotonin energy system, alleviate the depressive behavior of CRS mice.
