      [目的]探讨眼络通方对视网膜静脉阻塞(retinal vein occlusion,RVO)新西兰兔视网膜血管新生的影响。[方法]将48只健康新西兰兔随机分为空白对照组12只和模型组36只,模型组以氩激光光凝,建立RVO模型,并随机分为模型对照组、天保宁组、眼络通组,每组各12只。空白对照组及模型对照组予0.9%氯化钠溶液灌胃5mL/(kg·d);天保宁组灌胃给药剂量为0.02g/(kg·d);眼络通组灌胃给药剂量6.9g/(kg·d),连续给药4周。分别于灌胃治疗第1、2、4周后每组各处死4只新西兰兔,以免疫组化与Western blot法检测视网膜组织血小板源性生长因子(platelet-derived growth factor,PDGF)、血管内皮细胞生长因子(vascular endothelial growth factor,VEGF)的蛋白表达,实时荧光定量PCR(real-time quantitative polymerase chain reaction,RT-PCR)检测视网膜组织PDGF、VEGF的mRNA表达,并对相关数据进行统计学分析。[结果]与空白对照组比较,模型对照组1、2、4周各时相视网膜组织中PDGF、VEGF蛋白与mRNA表达量均显著升高(P<0.05);与模型对照组比较,眼络通组1、2、4周各时相视网膜组织中PDGF、VEGF蛋白与mRNA的表达量均显著降低(P<0.05),但仍显著高于空白对照组(P<0.05);而天保宁组各时相视网膜组织中PDGF、VEGF蛋白与mRNA的表达量差异无统计学意义(P>0.05)。[结论]眼络通方通过抑制视网膜组织VEGF、PDGF的表达,减少促血管新生因子释放,从而抑制视网膜血管新生,发挥保护视网膜、治疗RVO的作用。

      [Objective] To explore the effects of Yanluotong Decoction on retinal angiogenesis in retinal tissue of retinal vein occlusion(RVO) of New Zealand rabbits.[Methods] Forty-eight healthy New Zealand rabbits were randomly divided into 2 groups, including blank control group(n=12) and model group(n=36), RVO models were established by argon laser photocoagulation in model group,then RVO model rabbits were randomly divided into model control group, Tianbaoning group and Yanluotong group, 12 in each group. The blank control group and model control group were given 5mL/(kg·d) 0.9% sodium chloride solution; the Tianbaoning group was given 0.02g/(kg·d); the Yanluotong group was given 6.9g/(kg·d) corresponding drugs through intragastric administration for 4 weeks. Immunohistochemistry and Western blot were used to detect the protein expression of angiogenic factor platelet-derived growth factor(PDGF) and vascular endothelial cell growth factor(VEGF) in the retinal tissue of 4 New Zealand rabbits in each group after 1, 2, and 4 weeks of treatments. Real-time quantitative-polymerase chain reaction(RT-PCR) was used to detect the mRNA expression of PDGF and VEGF in the retinal tissue.[Results] Compared with blank control group,the protein and mRNA expression of PDGF, VEGF in retinal tissues of the model control group of 1, 2 and 4 weeks increased(P<0.05). Compared with model control group, the protein and mRNA expression of PDGF, VEGF in retinal tissues at all phases of week decreased in Yanluotong group(P<0.05), but still significantly higher than those of blank control group(P<0.05); and the expression levels of PDGF, VEGF protein and mRNA in retinal tissues of Tianbaoning group at all phases were not significantly different from model control group(P>0.05).[Conclusion] By inhibiting the expression of VEGF and PDGF in retina, Yanluotong Decoction can reduce the release of pro angiogenic factors, so as to inhibit retinal angiogenesis, protect retina and treat RVO.
