      [目的]通过分析扶正解毒合剂的有效化学成分,研究其治疗新型冠状病毒肺炎(corona virus disease 2019,COVID-19)的作用机制。[方法]通过中药系统药理学数据库与分析平台(Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform,TCMSP)对扶正解毒合剂的化学活性物质及作用于人体的靶点蛋白进行检索和筛选,同时应用基因组注释数据库平台(genome annotation database platform,Gene Cards)预测疾病的作用靶点,并运用Uniprot数据库查询与之相对应的基因名称,构建活性成分-靶点网络图,通过String数据库平台构建蛋白互作(protein-protein interaction,PPI)网络,再利用Bioconductor数据库和R语言进行基因本体(gene ontology,GO)富集分析和京都基因与基因组百科全书(Kyoto Encyclopedia of Genes and Genomes,KEGG)富集分析,构建靶点-通路网络图。[结果]通过筛选,共获得扶正解毒合剂作用于COVID-19的关键化学成分126个,共同靶点51个,通过PPI网络得到15个核心基因;GO富集分析结果包括脂多糖反应、膜筏、细胞因子受体结合等;KEGG富集通路包括精氨酸生物合成通路、精氨酸和脯氨酸代谢通路、花生四烯酸代谢通路。[结论]扶正解毒合剂可通过调节白细胞介素-2(interleukin-2,IL-2)、IL-6、丝裂原活化蛋白激酶(mitogen-activated protein kinase,MAPK)、核转录因子-κB p65(nuclear factor kappa B p65,NF-κB p65/RELA)、前列腺素内过氧化物合酶1(prostaglandin-endoperoxide synthase 1,PTGS1)、信号转导和转录激活子1(signal transducer and activator of transcription 1,STAT1)等靶点,调控晚期糖基化终产物及其受体(advanced glycation end products-receptor for advanced glycation end products,AGE-RAGE)信号通路、肿瘤坏死因子(tumor necrosis factor,TNF)信号通路、白介素-17(interleukin-17,IL-17)信号通路来抑制机体炎症反应,调节免疫功能,从而对COVID-19起到治疗作用。

      [Objective] To analyze the effective chemical constituents of Fuzheng Jiedu mixture and study its mechanism of action in the treatment of corona virus disease 2019(COVID-19).[Methods] Traditional Chinese Medicine System Pharmacology Database and Analysis Platform(TCMSP) database was used to search and screen the chemically active substances of Fuzheng Jiedu mixture and target proteins acting on the human body. At the same time, the genome annotation database platform(Gene Cards) was used to predict the targets of diseases. And use the Uniprot database to query the corresponding gene names, construct an active ingredient-target network diagram, construct a protein-protein interaction(PPI) network through the String database platform, and then use the Bioconductor database and R language Gene Oncology(GO) enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG) enrichment analysis to construct a target-path network diagram.[Results] Through screening, a total of 126 key chemical components of Fuzheng Jiedu mixture acting on COVID-19 and 51 common targets were obtained, and 15 core genes were obtained through PPI network. GO enrichment analysis results included response of lipopolysaccharide, membrane raft, cytokine receptor binding, etc; KEGG enrichment pathway included biosynthetic pathway, arginine and proline metabolism pathway, arachidonic acid metabolism pathway.[Conclusion] Fuzheng Jiedu mixture can inhibit the body's inflammatory response by regulating interleukin-2(IL-2), IL-6, mitogen-activated protein kinase(MAPK), nuclear factor kappa B p65(NF-κB p65/RELA), prostaglandin-endoperoxide synthase 1(PTGS1), signal transducer and activator of transcription 1(STAT1) and other targets, through the advanced glycation end products-receptor for advanced glycation end products(AGE-RAGE) signaling pathway, tumor necrosis factor(TNF) signaling pathway and IL-17 signaling pathway, it plays a therapeutic role in COVID-19 by inhibiting the body's inflammatory response and regulating immune function.
