[目的]利用网络药理学方法探究连花清瘟胶囊治疗新型冠状病毒肺炎(corona virus disease 2019,COVID-19)的作用机制。[方法]通过中药系统药理学分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)和人类基因组注释数据库(Human Genome Annotation Database,GeneCards)分别获取连花清瘟胶囊有效成分作用靶点和COVID-19靶点。利用String平台构建共同靶点的蛋白互作(protein-protein interaction,PPI)网络,并通过Cytoscape软件筛选核心靶点。在Metascape数据库进行基因本体(gene ontology,GO)和京都基因与基因组百科全书(Kyoto Encyclopedia of Genes and Genomes,KEGG)富集分析。运用Cytoscape构建“有效成分-核心靶点”网络,筛选主要有效成分。[结果]共获得连花清瘟胶囊有效成分184种,有效成分的作用靶点255个,COVID-19靶点587个,其中共同靶点65个。通过String平台及Cytoscape软件对共同靶点PPI网络进行拓扑学分析,得到血管内皮生长因子A(vascular endothelial growth factor A,VEGFA)、丝裂原活化蛋白激酶3(mitogen-activated protein kinase 3,MAPK3)、肿瘤抗原p53(tumor antigen p53,TP53)、白细胞介素-6(interleukin-6,IL-6)等23个核心靶点。Metascape数据库的富集分析表明,核心靶点主要涉及多种病毒感染通路。通过对“有效成分-核心靶点”进行交互网络分析,从连花清瘟胶囊中筛选得到槲皮素、木犀草素等22种主要活性成分,参与COVID-19的治疗。[结论]连花清瘟胶囊可以作用于VEGFA、MAPK3、TP53、IL-6等核心靶点,通过抑制炎症因子、调控心血管疾病相关通路、抗病毒等过程,发挥治疗COVID-19的作用。 |

[Objective] To investigate the mechanism of Lianhua Qingwen Capsule in the treatment of corona virus disease 2019(COVID-19) by network pharmacology. [Methods] Targets of effective components of Lianhua Qingwen Capsule and COVID-19 were obtained from the traditional Chinese medicine systems pharmacology database and analysis platform(TCMSP) and the Human Genome Annotation Database(GeneCards), respectively. The screening of common targets protein-protein interaction(PPI) network was carried out through the String platform and the core targets were screened by Cytoscape software. Gene ontology(GO) and Kyoto Encyclopedia of Genes and Genomes(KEGG) enrichment analysis were performed in Metascape database. Cytoscape was used to construct the “active components-core targets” network and screen the main active components. [Results] There are 184 active components in Lianhua Qingwen Capsule, and 255 targets of active components, 587 targets of COVID-19, and 65 common targets. By topology analysis of common target PPI network and String platform and Cytoscape software, 23 core targets including vascular endothelial growth factor A(VEGFA),mitogen-activated protein kinase 3(MAPK3),tumor antigen p53(TP53) and interleukin-6(IL-6) were obtained. Enrichment analysis by Metascape database showed that core targets were mainly involved in a variety of viral infection pathways. Twenty-two major components including quercetin and luteolin, which involved in the treatment of COVID-19, were analyzed through the “active component-core target” network. [Conclusion] Lianhua Qingwen Capsule could act on core targets such as VEGFA, MAPK3, TP53 and IL-6, and inhibit inflammatory factors, regulate cardiovascular disease related pathways and antiviral processes to achieve the therapeutic effect of COVID-19. |
