      [目的] 明确仙芎骨康颗粒对半月板-前交叉韧带联合损伤所致骨关节炎(osteoarthritis,OA)大鼠模型的治疗作用,并初步探索其作用机制。[方法] 48只SD大鼠随机分为空白对照组,模型组,仙芎骨康低、中、高剂量组,阳性对照组,除空白对照组外均采用Hulth法建立大鼠OA模型,造模与给药同时进行,仙芎骨康低、中、高剂量组分别灌胃给予仙芎骨康药液11.25、22.50、45.00 g·kg-1,阳性对照组灌胃给予盐酸氨基葡萄糖(0.09 g·kg-1)和塞来昔布(0.75 g·kg-1),模型组和空白对照组灌胃给予等量0.9%氯化钠注射液,连续给药8周。取膝关节组织制作病理切片,苏木精-伊红(hematoxylin-eosin,HE)染色和番红O(safranin O,SO)染色观察其病理变化;酶联免疫吸附试验检测血清中基质金属蛋白酶-13(matrix metallopeptidase-13,MMP-13)、白细胞介素-1β(interleukin-1β,IL-1β)水平。[结果] HE染色和SO染色显示各给药组大鼠膝关节软骨层明显增厚,软骨细胞增多,具有明显的修复效果,且仙芎骨康中、高剂量组、阳性对照组较仙芎骨康低剂量组改善效果更为明显。与空白对照组比较,模型组MMP-13、IL-1β水平升高(P<0.01)。与模型组比较,仙芎骨康中、高剂量组和阳性对照组MMP-13水平降低(P<0.01),三者组间差异无统计学意义(P>0.05),而低剂量组MMP-13水平与模型组差异无统计学意义(P>0.05)。与模型组比较,仙芎骨康低、中、高剂量组和阳性对照组IL-1β水平均降低(P<0.01),且给药组间差异无统计学意义(P>0.05)。[结论] 仙芎骨康颗粒具有防治OA的作用,其机制可能与下调MMP-13的表达、抑制炎症细胞因子IL-1β产生有关。

      [Objective] To clarify the effects and mechanism of Xianxiong Gukang particle on rats with osteoarthritis(OA) whose meniscus and anterior cruciate ligaments are injured. [Methods] Forty-eight SD rats were randomly divided into 6 groups, blank control group, model group, low, medium and high doses of Xianxiong Gukang groups and positive control group. Except blank control group, the model of OA was made according to the Hulth method. The modeling and administration were carried out simultaneously for 8 weeks. The rats in Xianxiong Gukang groups were orally administrated with different doses of Xianxiong Gukang particle 11.25, 22.50 and 45.00 g·kg-1, respectively; rats in positive control group were orally given hydrochloric acid amino glucose(0.090 g·kg-1) and celexib(0.75 g·kg-1); those in model group and blank control group were orally given an equal amount of 0.9% sodium chloride injection. The histological sections made from knee joint tissues were used to observe their pathological changes by hematoxylin-eosin(HE) and safranin O(SO) staining, and the serum levels of matrix metallopeptidase-13(MMP-13) and interleukin-1β(IL-1β) were detected by enzyme-linked immunosorbent assay(ELISA). [Results] The cartilage layers of the knee joint of rats in each treatment group were significantly thickened and chondrocytes were increased, which meant recovery, especially in medium and high doses of Xianxiong Gukang group and positive control group. Compared with blank control group, the serum levels of MMP-13 and IL-1β in model group increased(P<0.01). Compared with model group, MMP-13 in medium and high dose of Xianxiong Gukang groups and positive control group decreased(P<0.01), while there was no significant difference among the latter three groups(P>0.05); and there was no significant difference between low dose of Xianxiong Gukang group and model group(P>0.05). Compared with model group, IL-1β in the Xianxiong Gukang groups and positive control group decreased(P<0.01), while there was no significant difference among the latter four groups(P>0.05). [Conclusion] Xianxiong Gukang particle has the effect of preventing and treating OA, which probably relates to down-regulating expression of MMP-13 and inhibition of IL-1β secretion.
