      [目的] 探究肠道菌群在解毒祛瘀滋阴方治疗MRL/lpr狼疮小鼠过程中的作用。[方法] 将12周龄MRL/lpr小鼠随机分为模型组(0.9%氯化钠水溶液灌胃)、中药组(0.156 g·10 g-1解毒祛瘀滋阴方灌胃)和粪菌移植组(移植中药组小鼠的粪菌液),以12周龄C57BL/6小鼠作为空白组。饲养6周后采集小鼠血清样本,采用酶联免疫吸附试验(enzyme linked immunosorbent assay,ELISA)检测小鼠血清抗双链DNA抗体(anti-double stranded DNA antibody,anti-dsDNA)、抗核抗体(antinuclear antibody,ANA)和白细胞介素-6(interleukin-6,IL-6)表达水平;称量脾脏质量,计算脾脏指数;采用流式细胞术检测脾脏生发中心B细胞和浆细胞百分比;采用苏木精-伊红(hematoxylin-eosin,HE)染色分析肾脏病理变化。[结果] 与模型组比较,中药组和粪菌移植组的菌属Bacteroides和Sutterella丰度均显著上调(P<0.05),菌属Prevotella、Oscillospira、Ruminococcus、Odoribacter和Rikenella丰度均显著下调(P<0.05)。此外,解毒祛瘀滋阴方和粪菌移植均能显著降低MRL/lpr小鼠血清anti-dsDNA、ANA和IL-6水平(P<0.05,P<0.01),降低脾脏指数(P<0.05)和脾脏浆细胞百分比(P<0.05),减少肾脏炎性细胞浸润。[结论] 解毒祛瘀滋阴方可通过调控肠道菌群,发挥治疗系统性红斑狼疮的作用。

      [Objective] To explore the role of gut microbiota in the treatment of MRL/lpr mice by Jiedu Quyu Ziyin Prescription. [Methods] MRL/lpr mice of 12 weeks old were randomly divided into model group(oral gavage with 0.9% sodium chloride solution per day), traditional Chinese medicine group(oral gavage with 0.156 g·10 g-1 Jiedu Quyu Ziyin Prescription solution per day), and fecal microbiota transplantation(FMT) group(transplanted with the gut microbiota of traditional Chinese medicine group per day). In addition, C67BL/6 mice of 12 weeks old were set as the blank group. After six weeks of feeding, serum samples were collected to evaluate the expression levels of anti-double stranded DNA(anti-dsDNA) antibody, antinuclear antibody(ANA), and interleukin-6(IL-6); the spleen was weighed to calculate spleen index, the percentage of germinal center B cells and plasma cells in the spleen was detected by flow cytometry, and the renal pathology was analyzed by hematoxylin-eosin(HE) staining. [Results] Compared with model group, traditional Chinese medicine and FMT groups had a significant increase in genus Bacteroides and Sutterella(P<0.05), a significant decrease in genus Prevotella, Oscillospira, Ruminococcus, Odoribacter and Rikenella(P<0.05). Besides, both traditional Chinese medicine and FMT groups decreased levels of serum anti-dsDNA, ANA and IL-6(P<0.05, P<0.01), spleen index and the percentage of plasma cells in the spleen(P<0.05), and the infiltration of inflammatory cells in the kidney. [Conclusion] Jiedu Quyu Ziyin Prescription could play a role in the treatment of systemic lupus erythematosus by regulating gut microbiota.
