      [目的] 研究胃复春胶囊对人胃癌MKN-49P细胞的作用,为胃复春胶囊在抗消化道肿瘤方面的应用提供新的依据。 [方法] 受试样品为经胃复春胶囊饲喂大鼠的含药血清。胃癌MKN-49P细胞与不同浓度的胃复春胶囊含药血清共同孵育,采用噻唑蓝(methy thiazolyl tetrazolium,MTT)法及流式细胞术分别检测细胞活力和细胞凋亡情况;JC-1染色检测线粒体膜电位的变化;免疫印迹法检测信号传导与转录激活因子3(signal transducer and activator of transcription 3,STAT 3)蛋白及与细胞凋亡相关的蛋白表达情况。[结果] 胃复春胶囊含药血清能够剂量和时间依赖地抑制MKN-49P细胞的生长,并可诱导MKN-49P细胞凋亡。浓度为0~100 μg·mL-1的胃复春胶囊含药血清作用于MKN-49P细胞24 h后,细胞活力呈剂量依赖性降低;JC-1染色显示线粒体膜电位降低;经过25、50、100 μg·mL-1的胃复春胶囊含药血清分别处理24 h后,胃复春胶囊含药血清处理组细胞中STAT3的磷酸化减弱。[结论] 胃复春胶囊含药血清能抑制胃癌细胞的增殖并诱导其凋亡,其作用机制可能与STAT3信号通路有关,该结果为胃复春胶囊在临床上治疗胃癌提供了依据。

      [Objective] To study the effect of Weifuchun Capsule on human gastric cancer MKN-49P cells, and provide a new basis for the application of Weifuchun Capsule in anti-gastrointestinal cancer. [Methods] The test sample was the serum of rats fed with Weifuchun Capsule. Gastric cancer MKN-49P cells were incubated with different concentrations of drug-containing serum. Cell viability and apoptosis were detected by methy thiazolyl tetrazolium(MTT) assay and flow cytometry separated. JC-1 staining was applied to detect the changes of mitochondrial membrane potential(MMP). The expression levels of signal transducer and activator of transcription 3(STAT3) and apoptosis related proteins were detected by Western blot. [Results] Weifuchun Capsule containing serum could inhibit the growth of MKN-49P cells in a dose and time-dependent manner, and induce MKN-49P cells apoptosis. The cell viability of MKN-49P cells decreased in a dose-dependent manner after treated with 0~100 μg·mL-1 Weifuchun Capsule containing serum for 24 h. The MMP and phosphorylation of STAT3 were attenuated after treated with Weifuchun Capsule contained serum at the concentration of 25, 50, 100 μg·mL-1 for 24 h. [Conclusion] Weifuchun Capsule can inhibit the proliferation and induce apoptosis of gastric cancer cells, and its mechanism may be related to the STAT3 pathway. The present work provides a basis for the clinical treatment of gastric cancer with Weifuchun Capsule.
