      [目的] 探究芍药汤剂及颗粒剂成分的区别,以及对溃疡性结肠炎(ulcerative colitis,UC)的疗效差异。[方法] 采用超高效液相色谱-四级杆-时间飞行质谱仪联用技术(ultra-high-performance liquid chromatography-quadrupole/time of flight mass spectrometry,UPLC-Q-TOF/MS)检测两种剂型芍药汤样品,以MassLynx 4.1软件结合UNIFI软件分析结果。建立UC小鼠模型,随机分组,灌胃给药后,计算体质量下降率、疾病活动状态(disease activity index,DAI),以苏木精-伊红(hematoxylin-eosin,HE)染色观察结肠病理变化,原位末端转移酶标记(terminal-deoxynucleoitidyl transferase mediated nick end labeling,TUNEL)检测细胞凋亡率,酶联免疫吸附试验(enzyme-linked immunosorbent assay,ELISA)检测血清肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-10(interleukin-10,IL-10)含量。[结果] 芍药汤汤剂检出以小檗碱、芍药苷等为主的25种成分,颗粒剂检出以小檗碱、白杨黄素等为主的27种成分,两种剂型共同检出芍药苷、小檗碱等13种成分。与模型组比较,给药各组体质量下降率、DAI指数、病理学评分显著下降(P<0.001);结肠上皮细胞凋亡率显著下降(P<0.001,P<0.01),血清TNF-α含量显著下降(P<0.001),IL-10含量明显上升(P<0.05)。与颗粒剂比较,汤剂可明显降低体质量下降率(P<0.05)。[结论] 两种剂型芍药汤共同检出以芍药苷、小檗碱为主的13种成分,均对UC模型小鼠肠道炎症有明显改善作用;与颗粒剂相比,汤剂在改善小鼠体质量下降和保护结肠组织损伤方面疗效较好。

      [Objective] To explore the difference of components of Shaoyao Decoction and granules, and the therapeutic effect of Shaoyao Decoction on ulcerative colitis(UC). [Methods] Ultra-high performance liquid chromatography-quadrupole/time of flight mass spectrometry(PLC-Q-TOF/MS) was used to detect two dosage forms of Shaoyao Decoction, and MassLynx 4.1 software combined with UNIFI software was used to analyze the results. The UC mouse model was established, randomly grouped, and after administration by intragastric administration, the body mass decline rate and disease activity index(DAI) were calculated, and hematoxylin-eosin(HE) staining was used to observe the pathological changes of colon, terminal deoxynucleoitidyl transferase mediated nick end labeling(TUNEL) detected apoptosis rate, enzyme-linked immunosorbent assay(ELISA) was used to detect serum tumor necrosis factor-α(TNF-α) and interleukin-10(IL-10) content. [Results] Twenty-five components including berberine and paeoniflorin were detected in Shaoyao Decoction,27 components including berberine and populin were detected in granules, and 13 components including paeoniflorin and berberine were detected in both dosage forms. Compared with model group, the decline rate of body mass, DAI index and pathology score in all groups were significantly decreased(P<0.001); the apoptosis rate of colon epithelial cells was significantly decreased(P<0.001, P<0.01), the serum TNF-α content was significantly decreased(P<0.001) and the serum IL-10 content was significantly increased(P<0.05). Compared with granules, decoction could significantly reduce the decline rate of body mass(P< 0.05). [Conclusion] Two formulations of Shaoyao Decoction were used to detect 13 components mainly paeoniflorin and berberine, which can relieve intestinal inflammation of UC model mice obviously. Compared with granules, decoction has better curative effect on reducing body mass and protecting colonic tissue damage in mice.
